Moving from 16" to 18"

I'm moving from a 16" rim with 215/65/r16 tires to an 18" rim with 255/45/r18.

I know the tire will be a smaller profile, but will I need to get my speedometer recalibrated?

Look up the overall diameter of the tire on places like tirerack.com if they're the same then you won't need to correct it but if they aren't then you'll need to correct it for the new diameter.
